Description

BsaI is a type IIS restriction endonuclease commonly used for Golden Gate assembly and linearization of plasmids for mRNA vaccine production or other in vitro transcription purposes.

Enzyme Storage Buffer

10mM Tris-HCl, 200mM NaCl, 1mM DTT, 0.1mM EDTA, 200µg/ml BSA, 50% Glycerol, pH 7.4 @ 25°C.

1X BsaI Buffer

50mM Potassium Acetate, 20mM Tris-acetate, 10mM Magnesium Acetate, 100µg/ml BSA, pH 7.9 @ 25°C.

1X R Buffer

10mM Tris-HCl (pH 8.5 @ 37°C), 10mM MgCl2, 100mM KCl, 0.1mg/ml BSA.

Enzyme Digestion and Ligation Efficiency

When incubated with 20-fold excess of this endonuclease for 1 hour, >95% of the target fragments can be ligated and re-cut.

Activity Unit Definition

One unit is defined as the amount of enzyme required to digest 1µg of pXba DNA in 1 hour at 37°C in a total reaction volume of 50µl.

Store at -20°C.

Precautions

• The endonuclease should be stored in an icebox or on an ice bath during use, and after use, it should be immediately placed at -20°C for storage.
• The enzyme cutting activity can reach 100% at a temperature of 50°C.
• If the expected enzyme cutting site cannot be cleaved, please check for potential methylation interference. Dcm methylation overlapping with the enzyme cutting site can block the cleavage, and CpG methylation overlapping with the enzyme cutting site can also hinder cleavage.
• Glycerol content greater than 5% may cause asterisk activity.
• No asterisk activity is observed after a 3-hour incubation; delayed enzyme cutting may result in asterisk activity.
• This product is intended for use by professionals in scientific research and is not to be used for clinical diagnosis or treatment, food, or drugs, and should not be stored in ordinary residential areas.
