Blood Genomic DNA Isolation Kit with Magnetic Beads is a reagent kit that utilizes novel silica hydroxyl-coated magnetic beads as solid-phase media for the safe, convenient, stable, efficient, and high-quality purification of genomic DNA from animal blood.
The genomic DNA obtained after extraction using this kit can be used for various standard genomic DNA analyses and experimental assays, including PCR amplification of genomic DNA, restriction enzyme digestion, genotyping, Southern blotting, high-throughput sequencing (HTS), and construction of genomic DNA libraries.
Blood samples are rapidly lysed under the action of lysis buffer and proteinase K, releasing genomic DNA, which then specifically binds to the silica hydroxyl-coated magnetic beads under specific conditions. Subsequently, with the influence of an external magnetic field, such as a magnetic separation rack, the magnetic beads can be quickly and efficiently separated from the solution. After two washes to thoroughly remove non-specifically bound proteins, salts, and other impurities, genomic DNA is eluted from the magnetic beads using elution buffer, resulting in high-quality genomic DNA.
- This kit provides stable extraction results with a high yield and good purity. The genomic DNA extraction system of this kit has been repeatedly optimized, resulting in strong stability, high yield, and good purity. Approximately 4.5 µg of genomic DNA can be extracted from 100 µl of human whole blood, and approximately 5.6 µg of genomic DNA can be extracted from 100 µl of rabbit whole blood, with good purity typically reflected by an A260/A280 ratio of 1.7-1.9. This kit can extract high-quality genomic DNA from mammalian whole blood samples as low as 25 µl and anticoagulated samples with nucleated red blood cells as low as 5 µl.
- This kit offers convenience in its usage. The entire extraction process using this kit takes approximately 50 minutes at its fastest. On one hand, the kit optimizes the composition of the lysis buffer, allowing the lysis step to be completed in just 10 minutes. On the other hand, the extraction system is optimized, and the extraction step after lysis takes about 35 minutes. Compared to traditional centrifugation column-based extraction methods, this kit significantly simplifies the workflow and reduces the extraction time. When compared to similar magnetic bead purification products abroad, the required steps and operating time are essentially the same.
- This kit ensures safety in its usage. It employs special magnetic bead purification media for DNA separation and purification, eliminating the need for toxic and harmful organic reagents such as phenol and chloroform.
- This product is recommended for use with anticoagulated blood samples. For anticoagulated blood samples without nucleated red blood cells (e.g., human whole blood, mouse whole blood), a recommended sample volume is 100-200 µl. For anticoagulated blood samples containing nucleated red blood cells (e.g., aquatic animals, birds, poultry), a recommended sample volume is 5-15 µl. This kit is also suitable for extracting genomic DNA from cultured animal cells, with a recommended cell count of 1-2 million cells.
