Description
Traditional microRNA extraction kits are typically made using the principles of isothiocyanate/phenol/chloroform (TRIzol method) combined with high-concentration ethanol to adsorb short fragments of microRNA onto a silica membrane. However, the use of phenol/chloroform in this method is increasingly restricted due to its toxic and carcinogenic effects on the body and environmental concerns. Moreover, the applicability of the TRIzol method is limited, and for certain samples, including bacterial samples, the extraction of microRNA is not satisfactory.
Building on our advanced RNA/microRNA extraction technology, this kit innovatively adopts a technology pathway that doesn't rely on phenol or chloroform. Its unique universal lysis solution quickly lyses bacteria and inactivates bacterial RNA enzymes. Following this, the lysis mixture is passed through a genomic DNA removal column, where genomic DNA is removed while RNA (including microRNA) is selectively filtered. The filtered RNA/microRNA is then adjusted with ethanol to set the binding conditions and selectively adsorbed onto a silica membrane inside a centrifuge column under high salt conditions. After a series of quick washing and centrifugation steps using special washing solutions, cellular metabolites and proteins are removed. Finally, pure RNA (including microRNA) is eluted from the silica membrane with low-salt RNase-free H2O.
