Description
Arctiin (NSC 315527), a plant lignan extractable from Arctium lappa (burdock) seeds, is a potential environmental endocrine disruptor compound. It has been demonstrated to affect sex hormone metabolism, protein synthesis, and steroid biosynthesis.

In Vitro Study
Treatment of PC-3 cells with arctiin resulted in a concentration- and time-dependent decrease in cell number under serum-containing conditions. Arctiin primarily induced cell detachment without exhibiting anti-proliferative or cytotoxic effects in PC-3 cells. The arctiin-induced effect was found to be inhibited by cycloheximide, suggesting a requirement for protein synthesis. While arctiin, the active component of AL described in the literature, did not reduce degranulation in RBL-2H3 cells, a single high-performance liquid chromatography (HPLC) fraction from the AL extract showed inhibition of beta-hexosaminidase release (IC50= 22.2 microg/ml). The growth inhibition attributed to arctiin is linked to the down-regulation of cyclin D1 protein expression. Moreover, the arctiin-induced suppression of cyclin D1 protein expression is observed across various types of human tumor cells, including osteosarcoma, lung, colorectal, cervical, and breast cancer, melanoma, transformed renal cells, and prostate cancer. Depletion of the cyclin D1 protein using small interfering RNA rendered human breast cancer MCF-7 cells insensitive to the growth inhibitory effects of arctiin, implicating cyclin D1 as an important target of arctiin.
In Vivo Study
Histopathological evaluation of the prostate revealed that all rats in each group developed adenocarcinoma in the dorsolateral lobe of the prostate, except for two rats in the 0.1% arctiin-treated group and one rat in the 0.002% arctiin-treated group, which did not develop prostate adenocarcinoma. Following oral administration of arctiin (30, 60, 120 mg/kg/day) for three weeks, there was a significant decrease in the levels of serum creatinine (Scr) and blood urea nitrogen (BUN), as well as a marked reduction in 24-hour urine protein content, while the endogenous creatinine clearance rate (ECcr) significantly increased. STZ-induced diabetic rats were treated with arctiin at dosages of 60 or 40 mg/kg/day via intraperitoneal injection for 8 weeks. Blood glucose and 24-hour urinary albumin content were measured, and kidney histopathological changes were monitored.
