Description
Anti-His Magnetic Beads, also known as Anti-HHHHHH Magnetic Beads, Anti-His Immunomagnetic Beads, or His Antibody Magnetic Beads, are formed by covalently coupling high-quality mouse monoclonal antibodies against the His tag with nano-sized amino magnetic beads. They can specifically bind to proteins containing the His tag in lysates, serum, ascites, and other samples derived from animals, plants, or microorganisms. These beads are used for immunoprecipitation (IP), co-immunoprecipitation (Co-IP), or purification of fusion proteins or protein complexes carrying the His tag.
His tag, Flag tag, Myc tag, HA tag, His tag, GST tag, and other commonly used tags are frequently employed in expression vectors. Fusion expression with these tags allows convenient detection of the target protein and its interacting partners and facilitates the purification of the target protein.
The His tag is a peptide composed of six consecutive histidine residues (HHHHHH). By using genetic recombination techniques, the nucleotide sequence encoding the His tag is connected to the 5' or 3' end of the target gene to express the desired protein fused with the His tag. The His tag has the following advantages: it has a very small molecular weight, only 0.84 kDa, and usually does not interact with the target protein or affect its function. It typically does not form dimers and does not interfere with downstream applications of the protein. As a tag protein, subsequent detection, localization, and functional analysis of the target gene can be performed using His antibodies and Anti-His Magnetic Beads. Protein purification using the His tag is simple, and purification conditions are mild. In many cases, the His tag does not need to be removed after protein purification and does not affect the protein's function. The His tag can be applied to various protein expression systems and can be combined with other affinity tags to construct dual-affinity tag expression systems. The N-terminal His tag is compatible with bacterial transcription-translation mechanisms, which is advantageous for protein expression. Based on these advantages, the His tag has been widely used in various aspects of protein expression, purification, identification, interaction studies, and functional analysis. While the purification of His-tagged proteins is commonly performed using His-tagged protein purification media (nickel columns), Anti-His Magnetic Beads offer a simpler and more convenient approach for the purification or immunoprecipitation of fusion proteins or protein complexes carrying the His tag, especially for small-scale purifications or immunoprecipitation experiments.
Anti-His Magnetic Beads specifically bind to fusion proteins carrying the His tag and can be conveniently applied in immunoprecipitation, purification, and other experiments involving fusion proteins or protein complexes with the His tag, using magnetic separation devices such as magnetic racks.
