Animal RNA Isolation Kit with Magnetic Beads is a reagent kit that utilizes novel nucleic acid purification media-coated magnetic beads. It allows for the safe, convenient, stable, efficient, and high-quality extraction of total RNA from animal tissues or cultured animal cells. The extracted RNA includes both high-molecular-weight RNA and small RNA (<200nt), often referred to as Total RNA, suitable for various molecular biology and biochemical applications.
The RNA obtained with this kit can be used for downstream experiments such as reverse transcription, RT-PCR, qPCR, Northern blotting, dot blotting, mRNA purification, in vitro translation, RNase protection assays, cDNA cloning, among others. It is also suitable for applications with high RNA quality requirements, such as gene expression microarray analysis and high-throughput sequencing.
RNA in animal tissues or cells can be categorized by length into long RNA (typically >200nt) and small RNA (usually <200nt). Long RNA can further be divided into long non-coding RNA (lncRNA) and mRNA, depending on whether it encodes proteins or peptides. Small RNA includes non-coding RNAs like 5.8S ribosomal RNA (rRNA), 5S rRNA, transfer RNA (tRNA), microRNA (miRNA), small interfering RNA (siRNA), Piwi-associated small RNA (piRNA), tRNA-derived small RNA (tsRNA), and small rDNA-derived RNA (srRNA).
In the kit, samples are rapidly lysed in the lysis solution, releasing total RNA. The RNA then specifically binds to the magnetic beads and is thoroughly purified through three washing steps to remove non-specifically bound proteins, salts, and other impurities. Finally, high-purity RNA is eluted using the elution buffer.

- This kit offers safe usage. It utilizes a specialized magnetic bead purification medium for RNA separation and purification, effectively avoiding the use of toxic and harmful organic reagents like phenol and chloroform, which are commonly used in traditional Trizol-based RNA extractions.
- The kit provides a convenient workflow. Employing magnetic bead purification, it eliminates the need for cumbersome RNA precipitation steps, reducing the extraction process to just 15-20 minutes. Compared to traditional Trizol-based extraction methods, this kit streamlines the procedure, shortening extraction time, and lowering the risk of RNA degradation. When compared to similar magnetic bead purification products, the required steps and processing times are generally consistent.
- The kit delivers high RNA yields. Its extraction efficiency has been thoroughly tested, with approximately 15-20µg of RNA extracted from 1 million HeLa cells, around 25-35µg of RNA extracted from 20mg of mouse liver tissue, and approximately 10-20µg of RNA extracted from 20mg of mouse brain tissue (higher yields are obtained from fresh samples compared to frozen samples).
Apart from DNase and Reaction Buffer (10X), store at room temperature, with a shelf life of one year. DNase and Reaction Buffer (10X) should be stored at -20ºC, with a shelf life of one year. RNA magnetic beads can be stored at 4ºC for extended periods when not in use, and storing them at 4ºC allows for longer-term preservation.
- Self-provided magnetic separation equipment is required.
- For samples rich in RNase, such as animal tissues, it is advisable to add Dithiothreitol (DTT) to the lysis solution to a final concentration of 40mM (e.g., add 20µl of 2M DTT to 1ml lysis solution) or β-Mercaptoethanol to a final concentration of 1% (e.g., add 10µl of β-Mercaptoethanol to 1ml lysis solution). Lysis solutions containing DTT or β-Mercaptoethanol can be stored at room temperature for up to one month.
- If DNase I provided in the kit is not used for treatment, the RNA extracted with this kit may contain a small amount of DNA. For downstream experiments sensitive to residual DNA, it is recommended to add an appropriate amount of DNase I to the magnetic beads after following the instructions in step 4. Please refer to the user manual for specific instructions.
- We recommend using the recommended number of cells or tissue quantity for RNA extraction; otherwise, it may cause aggregation of BeyoMag™ RNA magnetic beads, resulting in incomplete digestion of genomic DNA. If genomic DNA digestion is incomplete, you can increase the amount of DNase I or extend the incubation time.
- All reagents provided in this kit are RNase-free. Handle with care to avoid RNase contamination. All self-prepared reagents and consumables should also be RNase-free. If consumables may be contaminated with RNase, consider soaking them in 0.01% DEPC-treated water overnight, then autoclaving and drying at high temperature and pressure. Avoid breathing or talking over samples or the kit consumables to prevent RNase contamination. We recommend wearing disposable masks during operations.
- For the removal of RNAse in the laboratory environment, it is recommended to useRNase and DNase Away to remove RNase from laboratory surfaces or other contact surfaces.
- The effectiveness of RNA extraction from frozen cells or tissues is usually slightly lower than that from fresh cells or tissues. This is because some RNase inside cells or tissues can be released during the freeze-thaw process, leading to partial degradation of RNA in the samples.
- All operations for this kit are performed at room temperature, and there is no need for ice baths.
- The binding solution and washing solution contain a certain concentration of ethanol. After use, tighten the bottle caps to prevent evaporation and follow the relevant regulations for the storage and use of flammable reagents.
- This product is for scientific research by professionals only and should not be used for clinical diagnosis or treatment, food, or drugs. It should not be stored in a regular residential area.
