Alkaline Phosphatase (Fast) catalyzes the hydrolysis of DNA, RNA, and nucleotide 5' and 3' phosphate groups, as well as the removal of protein phosphate groups. However, it does not catalyze the hydrolysis of phosphodiester or phosphotriester bonds. Under 37°C conditions for 10 minutes, this enzyme can dephosphorylate the ends of all types of DNA. Since it retains 100% activity in RapidCleave™ restriction enzyme buffer and is inactivated by incubation at 80°C for 20 minutes, it can be combined in the same tube with RapidCleave™ fast restriction enzyme for a "digest-dephosphorylate" reaction, greatly simplifying the experimental process.

Unit Definition

Under conditions with Alkaline Phosphatase buffer at 37°C, the amount of enzyme required to dephosphorylate the 5' ends of 1 μg of linearized pUC57 DNA within 10 minutes is defined as one unit (U) of activity.

 

Quality Control

  • Residual Endonuclease Activity Detection: Incubate the enzyme solution with supercoiled plasmid DNA at 37°C for 4 hours. Analyze by DNA electrophoresis to confirm no change in the plasmid.
  • Residual Exonuclease Activity Detection: Incubate the enzyme solution with double-stranded DNA substrate at 37°C for 16 hours. Analyze by DNA electrophoresis to confirm no change in the double-stranded DNA substrate.
  • Host Cell Detection: Detect residual nucleic acids in the enzyme solution using E. coli 16S rDNA-specific Taq Man qPCR. E. coli genomic residues are below 10 copies.
