Description
Alkali-Resistant Recombinant Protein A, also known as Alkali-Tolerant Protein A, is a genetically engineered variant derived from natural Protein A. Through targeted screening and modification, this novel protein exhibits enhanced alkali resistance, improved stability, and stronger affinity, with increased specificity for IgG binding.
This product has been refined to remove the cell wall, membrane, and albumin-binding domains found in natural Protein A, retaining only the IgG-binding domains to ensure highly specific interactions. It demonstrates binding capabilities across various species’ IgG comparable to native Protein A, while withstanding prolonged exposure to 0.5 M NaOH, making it ideal for coupling with magnetic beads or agarose matrices as a ligand.
The alkali-resistant Protein A beads or agarose matrices tolerate in situ cleaning or sanitation with 0.1–0.5 M NaOH, effectively reducing the risk of endotoxin contamination and cross-contamination between batches. This resilience helps extend the lifespan of purification media and makes it especially well-suited for large-scale purification of monoclonal antibodies and therapeutic Fc-fusion proteins at the industrial level.
Compared to conventional Protein A, the alkali-resistant variant offers superior alkali tolerance. After 160 hours of exposure to 0.5 M NaOH, it retains nearly full antibody-binding capacity, whereas standard Protein A shows a reduction to 53% of its binding activity after just 48 hours under the same conditions.

Applications
Alkali-resistant Recombinant Protein A is ideal for coupling as a ligand to magnetic beads or agarose matrices.
