Description

Ac4ManNAz, also known as N-azidoacetylmannosamine-tetraacylated, is a specialized chemical reagent used in glycosylation labeling. It is an azide-containing metabolic glycoprotein labeling compound that can be taken up by cells and selectively modifies proteins. This reagent is commonly utilized for cell labeling, tracking, and proteomic analysis.

Ac4ManNAz contains an azide group, which allows it to undergo copper-catalyzed azide-alkyne cycloaddition (CuAAC) reactions with molecules containing alkyne groups. It can also participate in strain-promoted alkyne-azide cycloaddition (SPAAC) with molecules containing DBCO or BCN groups. These reactions make it a versatile tool for bioorthogonal labeling and visualization of glycoconjugates.

The compound is cell-permeable and intracellularly processed, replacing its natural counterpart, N-acetylmannosamine (ManNAc). This substitution results in azide-functionalized glycoconjugates that can be detected using click chemistry techniques. Researchers often use Ac4ManNAz for tasks such as introducing biotin groups for purification or fluorescent groups for microscopic imaging.

Ac4ManNAz is available in various quantities and is typically stored at -20°C to maintain its stability. It is soluble in solvents like DMSO, DMF, and methanol, and appears as a white to off-white solid. Its molecular formula is C₁₆H₂₂N₄O₁₀, with a molecular weight of 430.37 g/mol.
