Description
9N2 DNA Ligase catalyzes the formation of phosphodiester bonds, joining the 5´-phosphate and 3´-hydroxyl termini of two adjacent oligonucleotide strands hybridized to the same complementary target DNA strand.
This enzyme is derived from an extremely thermophilic deep-sea archaeon (Thermococcus sp.), specifically from a ligase gene cloned from the 9°N strain. It remains active in a temperature range of 45°C to 70°C. Through genetic engineering, its thermal stability has been further enhanced, allowing it to retain nearly full activity even after a 5-minute incubation at 95°C.

Activity Definition
One unit of 9N2 DNA Ligase is defined as the amount of enzyme required to ligate 50% of 1 pmol of a 12 bp cohesive-end substrate in a 20 μl reaction system at 50°C within 15 minutes.
