Description
3173 DNA Polymerase is a thermostable DNA polymerase derived from a virus. In addition to its DNA-dependent polymerase activity, it also has RNA-dependent reverse transcriptase activity, making it suitable for single-tube RT-PCR amplification. This enzyme has strong strand displacement activity and can be used for isothermal amplification of DNA or RNA. It also has 3’-5’ exonuclease activity, which can increase the fidelity of DNA synthesis but reduces synthesis efficiency. In LAMP experiments using RNA as a template, it can achieve single-enzyme system reactions. For RNA templates with complex secondary structures, heating at 92°C for 3 seconds can yield good amplification results, whereas heating Bst pol at this temperature leads to loss of amplification activity.
This enzyme is paired with two types of buffers: 10×3173 Isothermal Buffer and 10×AM PCR Buffer. The 10×3173 DNA Polymerase Buffer is used for isothermal PCR amplification, with a reaction temperature not exceeding 70°C. The 10×AM PCR Buffer is used for conventional PCR amplification, with a pre-denaturation time at 94°C not exceeding 5 minutes.

Activity Definition
One unit of activity is defined as the amount of enzyme required to incorporate 25 nmol of dNTP into acid-insoluble material in 30 minutes at 65°C in a 1× 3173 DNA Polymerase reaction buffer system.
Activity Assay Conditions
Incubate at 65°C in a 1× 3173 DNA Polymerase reaction buffer system for 30 minutes.
