Description
0.2% Alizarin Red S Staining Solution, pH8.3 is a convenient and rapid staining solution used for detecting calcium salt deposits, suitable for staining intracellular calcium deposits in cells.
Calcium is the most abundant inorganic salt in animals and constitutes a major mineral component of bones and teeth. It also plays a crucial role in various physiological processes such as muscle contraction and nerve transmission. In normal conditions, calcium does not appear in solid form in tissues outside of bones and teeth. However, in pathological conditions, calcium can precipitate as calcium salts like calcium carbonate and calcium phosphate, accumulating within tissues.
Alizarin Red S staining is a commonly used method widely applied in studies related to osteoblast differentiation, bone cell research, and pathological investigations. Alizarin Red S (ARS), also known as Alizarin Red sodium sulfate, Alizarin Red lake, has a CAS number of 130-22-3, a molecular formula of C14H7NaO7S, and a molecular weight of 342.25. Alizarin Red S is an anthraquinone derivative that can form stable orange-red or deep red complexes by chelating with various metal ions. It serves as a good indicator for metals and can be used as a colorimetric agent for determining metal ions using absorbance methods, an acid-base indicator, and also in polarographic analysis for metal ions. In histology and pathology, Alizarin Red S is often used to stain or locate calcium deposits in tissues, as it can chelate with calcium salts such as calcium carbonate or calcium phosphate, resulting in orange-red complexes.
The concentration of Alizarin Red S in this product is 0.2%, with a pH of 8.3, rendering it alkaline in nature. This staining solution is suitable for staining intracellular calcium deposits, such as vascular calcification, muscle calcification, ectopic calcification, and identifying calcium deposits in cultured cells. Since intracellular calcium deposits are usually limited in quantity, acidic staining solutions can potentially dissolve calcium, leading to poor staining results or false negatives. Hence, alkaline staining solutions are generally employed. Additionally, alkaline staining solutions often allow for longer staining times, yielding more satisfactory staining outcomes.
Calculating based on using 100µl of staining solution per slide, the 100ml package of this product can be used to stain 1000 slides.
