SELEX Aptamer Library Screening
Description
SELEX (Systematic Evolution of Ligands by Exponential Enrichment) is a technique used to screen highly specific nucleic acid aptamers from a random single-stranded nucleic acid sequence library. The fundamental concept of SELEX is to chemically synthesize an in vitro single-stranded oligonucleotide library, mix it with the target substance, and through repeated rounds of selection and amplification, wash away nucleic acid molecules that do not bind or have low affinity for the target substance, thereby isolating and purifying nucleic acid aptamers with high affinity for the target substance.
SELEX technology and nucleic acid aptamers offer several advantages, including a large library capacity, broad applicability, high resolution, strong practicality, high affinity, relatively simple, fast, and economical screening processes, and small aptamer size. This technology has applications in in vitro diagnostics, in vivo therapy, and drug development.
Client Provided Samples
Applicable Targets for SELEX Aptamer Screening Services: Proteins, small molecules, cells, bacteria, etc.
Protein Sample Requirements: Please provide the protein size, volume, concentration, buffer composition, protein gel image, and information on whether the protein contains any tags and the corresponding tag details.
- Proteins should not contain large tags.
- Buffers should not contain free amines, such as sodium azide.
- Tris buffer should not be used, and proteins should be stable in PBS buffer.
- Purity should be greater than 95% (recommended >95%, minimum 90%).
- Total protein amount should be 500 μg to 1 mg, with a concentration of 1 mg/ml.
Small Molecule Sample Requirements: Please provide the sample mass, concentration, volume, and storage conditions.
- Good water solubility (≥1 mM).
- Stability: resistant to oxidation and lactonization, etc.
- Minimal heat of dilution.
- Small molecule purity >95% (recommended >95%, minimum 90%).
- Sample amount: 10 mg.
Special Note: It is recommended to send small molecules in dry powder form. They can be transported with ice packs, or dry ice for added safety, depending on the storage requirements of the small molecules.
For Cells and Bacterial Targets: Please consult for specific sample requirements and quantities before sending.
Delivery
Final Product Delivery:
- Five aptamer sequences selected by the client
Delivery Report:
- NGS sequencing results
- Complete experimental report (including experimental procedures, enrichment rate data during the screening process, related photos, etc.)
Related Projects
Project:
- Aptamer Affinity Characterization Service
Project Description:
- Affinity characterization service supporting the aptamer screening service, primarily for determining the affinity between the target (protein or small molecule) and the aptamer. Specific charges need to be consulted.
