Description
PrimeIAmp™ HotStart LAMP Bst P L-HNB MasterMix is a 4× semi‑premixed solution that can be directly used for lyophilization experiments without any additional auxiliary reagents. The premix contains reaction buffer, dNTPs, Mg²⁺, HotStart Bst P polymerase, trehalose, mannitol, and other stabilizers.
L-HNB dye in the product combines with Mg2+ to produce deep violet color before the reaction starts. With the LAMP reaction, the resulting PPi (inorganic pyrophosphate) combines with Mg2+ in the system, making L-HNB show light green.

Features
Bst P DNA/RNA System has the following performance:
- The whole Bst P DNA/RNA System includes hot start Aptamer, which ensures that the enzyme activity blocking efficiency is >95% at <30°C, and the enzyme activity is completely released within 1 min at >60°C. This characteristic facilitates the establishment of the reaction system at room temperature and greatly reduces the non-specific amplification at low temperatures.
- The reaction temperature is further raised to 70°C, which greatly reduces the formation of primer dimer, improves the amplification specificity, and makes the nucleic acid release of crude samples more sufficient.
- The whole portfolio contains Helicase, so Premium LAMP amplification (pLAMP) is allowed without using F3/B3 primers. At the same time, Helicase has the function of assisting in strand unwinding, which further reduces the concentration of FIP/BIP primers. This will further reduce non-specific amplification and greatly improve amplification homogeneity.

Storage
For long-term storage, keep the product at –20 °C or below (stable for up to 18 months). The reagent can withstand up to 10 freeze–thaw cycles without performance loss, though repeated freeze–thawing should be avoided when possible. For short-term use, storage at 2–8 °C is recommended, where the product remains stable for up to 2 weeks. Due to the high concentration of lyophilization excipients, a small amount of white insoluble material may be observed upon thawing; this will fully dissolve after 3–5 minutes at 30 °C (or by warming in the palm), without affecting product performance.
Storage
Store at –20 °C or below for up to 24 months. The product typically melts within 3–5 minutes at 28 °C, and up to 20 freeze–thaw cycles do not affect performance.
